Cyro-electron microscopy of tau filaments from people with corticobasal degeneration reveals a previously unseen four-layered fold, distinct from the filament structures seen in Alzheimer'  s disease, Pick'  s disease and chronic traumatic encephalopathy.

Corticobasal degeneration (CBD) is a neurodegenerative tauopathy-a class of disorders in which the tau protein forms insoluble inclusions in the brain-that is characterized by motor and cognitive disturbances(1-3). The H1 haplotype of MAPT (the tau gene) is present in cases of CBD at a higher frequency than in controls(4,5), and genome-wide association studies have identified additional risk factors(6). By histology, astrocytic plaques are diagnostic of CBD7,8  by SDS-PAGE, so too are detergent-insoluble, 37 kDa fragments of tau(9). Like progressive supranuclear palsy, globular glial tauopathy and argyrophilic grain disease(10), CBD is characterized by abundant filamentous tau inclusions that are made of isoforms with four microtubule-binding repeats(11-15). This distinguishes such '  4R'  tauopathies from Pick'  s disease (the filaments of which are made of three-repeat (3R) tau isoforms) and from Alzheimer'  s disease and chronic traumatic encephalopathy (CTE) (in which both 3R and 4R isoforms are found in the filaments)(16). Here we use cryo-electron microscopy to analyse the structures of tau filaments extracted from the brains of three individuals with CBD. These filaments were identical between cases, but distinct from those seen in Alzheimer'  s disease, Pick'  s disease and CTE17-19. The core of a CBD filament comprises residues lysine 274 to glutamate 380 of tau, spanning the last residue of the R1 repeat, the whole of the R2, R3 and R4 repeats, and 12 amino acids after R4. The core adopts a previously unseen four-layered fold, which encloses a large nonproteinaceous density. This density is surrounded by the side chains of lysine residues 290 and 294 from R2 and lysine 370 from the sequence after R4.

A synthetic approach is described, for efficiently converting non-van der Waals solids into two-dimensional van der Waals transition-metal chalcogenide layers with specific phases, enabling the high-throughput production of monolayers.

Although two-dimensional (2D) atomic layers, such as transition-metal chalcogenides, have been widely synthesized using techniques such as exfoliation(1-3) and vapour-phase growth(4,5), it is still challenging to obtain phase-controlled 2D structures(6-8). Here we demonstrate an effective synthesis strategy via the progressive transformation of non-van der Waals (non-vdW) solids to 2D vdW transition-metal chalcogenide layers with identified 2H (trigonal prismatic)/1T (octahedral) phases. The transformation, achieved by exposing non-vdW solids to chalcogen vapours, can be controlled using the enthalpies and vapour pressures of the reaction products. Heteroatom-substituted (such as yttrium and phosphorus) transition-metal chalcogenides can also be synthesized in this way, thus enabling a generic synthesis approach to engineering phase-selected 2D transition-metal chalcogenide structures with good stability at high temperatures (up to 1,373 kelvin) and achieving high-throughput production of monolayers. We anticipate that these 2D transition-metal chalcogenides will have broad applications for electronics, catalysis and energy storage.

Solar heating of a cometary surface provides the energy necessary to sustain gaseous activity, through which dust is removed(1,2). In this dynamical environment, both the coma(3,4) and the nucleus(5,6) evolve during the orbit, changing their physical and compositional properties. The environment around an active nucleus is populated by dust grains with complex and variegated shapes(7), lifted and diffused by gases freed from the sublimation of surface ices(8,9). The visible colour of dust particles is highly variable: carbonaceous organic material-rich grains(10) appear red while magnesium silicate-rich(11,12) and water-ice-rich(13,14) grains appear blue, with some dependence on grain size distribution, viewing geometry, activity level and comet family type. We know that local colour changes are associated with grain size variations, such as in the bluer jets made of submicrometre grains on comet Hale-Bopp(15) or in the fragmented grains in the coma(16) of C/1999 S4 (LINEAR). Apart from grain size, composition also influences the coma'  s colour response, because transparent volatiles can introduce a substantial blueing in scattered light, as observed in the dust particles ejected after the collision of the Deep Impact probe with comet 9P/Tempel 1(17). Here we report observations of two opposite seasonal colour cycles in the coma and on the surface of comet 67P/Churyumov-Gerasimenko through its perihelion passage(18). Spectral analysis indicates an enrichment of submicrometre grains made of organic material and amorphous carbon in the coma, causing reddening during the passage. At the same time, the progressive removal of dust from the nucleus causes the exposure of more pristine and bluish icy layers on the surface. Far from the Sun, we find that the abundance of water ice on the nucleus is reduced owing to redeposition of dust and dehydration of the surface layer while the coma becomes less red.
